Master thesis project: Development of alternative analytical methods for quantification of excipients
Formulations of protein-based pharmaceuticals often include surfactants to help stabilizing the proteins and maintaining their functionality. Examples are PS20, PS80, PX188 and FM1000. During development and production of protein-based pharmaceuticals it is important to quantify the content of protein as well as excipient. However, separation between these surface-active excipients (surfactants) and the protein can be challenging and lead to misleading analysis results.
At present, chromatographic methods with different separation principles are used, e.g. reverse-phase (RP) and size-exclusion (SE). These are combined with different detection principles, e.g. CAD, QDa/MS. In this project, alternative methods for quantitative determination of excipients will be explored and tested. This may include further development of existing chromatographic methods as well as combining chromatographic methods with new analysis principles.
Project scope• Development of methods for quantitative determination of excipients such as surfactants that help stabilizing the proteins and maintaining their functionality.• Explore chromatographic methods with different separation principles, e.g. reverse-phase (RP) and size-exclusion (SE), combined with different detection principles, e.g. CAD, QDa/MS for excipient analysis.• Establishment of a method by which excipients can be robustly quantified
Desired skills/compentencies• Solid knowledge within chemical analysis, preferably chromatography• Interest in biophysics and surface chemistry• Practical laboratory skills
